She was the eldest child and daughter of Anton Ulrich, Duke of Saxe-Meiningen and his second wife, Landgravine Charlotte Amalie of Hesse-Philippsthal. Charlotte was an elder sister of Charles William, Duke of Saxe-Meiningen and George I, Duke of Saxe-Meiningen .
Hermann was the third son of Prince Bernard of Saxe-Weimar-Eisenach (1792-1862) from his marriage to Ida of Saxe-Meiningen (1794–1852), the daughter of George I, Duke of Saxe-Meiningen.
