De Niro é filho do pintor americano Robert De Niro, Sr., morto em 93. Foi casado com a comissária de voo Grace Hightower de 1997 a 2018 De Niro tem sete filhos. De Diahnne Abbott tem dois, Drena e Raphael. Em 1995 teve os gémeos Julian e Aaron com a ex-namorada, a modelo e atriz Toukie Smith.
