Dorothy Stafford, Lady Stafford (1 October 1526 – 22 September 1604) was an English noblewoman, and an influential person at the court of Queen Elizabeth I of England, to whom she served as Mistress of the Robes. Dorothy Stafford was the second wife of Sir William Stafford, widower of Mary Boleyn.

Dorothy Stafford of Penthurst, Kent (b. c. 1526 - d. 22 Sep 1604), was the daughter of Henry de Stafford and Ursula de la Pole (c. 1504-70). She's a direct descendant of the dukes of Buckingham. Dorothy's paternal grandfather Sir Edward Stafford, 3rd Duke of Buckingham (03 Feb 1478 – 17 May 1521), was executed by Henry VIII.
