Mackenzie emigrated in 1842 from Scotland to Canada West (now Ontario), where he worked as a stone mason and established himself as a building contractor at Sarnia.

He led a party of nine men on a 2,800-mile round-trip dash from Canada to the Pacific Ocean, using rivers, native guides and his own navigation skills. He also published a book about his voyage and urged British ministers to establish ports for fur trade.
