O príncipe Guilherme de Saxe-Weimar-Eisenach nasceu a 21 de Dezembro de 1853 em Estugarda. Era o filho mais velho do príncipe Hermano de Saxe-Weimar-Eisenach e da princesa Augusta de Württemberg (1826-1898).

Nascido em Weimar, era o filho mais velho de Carlos Augusto, Grão-Duque Hereditário de Saxe-Weimar-Eisenach, herdeiro do grão-duque, e da princesa Paulina de Saxe-Weimar-Eisenach. Sucedeu ao seu avô, Carlos Alexandre, como grão-duque a 5 de Janeiro de 1901, uma vez que o seu pai tinha já falecido.

Saxe-Weimar (German: Sachsen-Weimar) was one of the Saxon duchies held by the Ernestine branch of the Wettin dynasty in present-day Thuringia. The chief town and capital was Weimar. The Weimar branch was the most genealogically senior extant branch of the House of Wettin.
