Sarah Lois Vaughan est née le 27 mars 1924 à Newark (New Jersey), fille d’Asbury et d’Ada Vaughan. Dans sa jeunesse, elle prend des leçons de piano et fréquente l’église baptiste de Mount Zion, où elle est claviériste.
