Beliau dilahirkan di Moscow, Empayar Rusia pada 13 Mac 1913. Pada tahun 1942, Mikhalkov bekerja melukis atas perhatian Kesatuan Soviet. Pada tahun 1944, Sergei bekerja dengan Alexander Vasilyevich Alexandrov untuk menulis lirik lagu kebangsaan mereka. Ia tambah teruk apabila Alexander meninggal dunia pada 1946 kerana tuberkulosis.
